hsa-mir-21_DDSD (BBa_K5086012) is a single-stranded DNA sequence with 70 nucleotides that is a self-enhancing probe for the target hsa-mir-21_Rep. The sequence at its 3' end is designed to be complementary to the Universalness_h strand, so that it binds specifically to the Universalness_h strand and thus accomplishes recognition of the input signal. DNA polymerase polymerizes DNA in the 5'-3' direction from the 3' end of the target hsa-mir-21_Rep. During DNA polymerization by Bst DNA polymerase, the wt strand is replaced and released by the newly synthesized daughter strand, and a single strand of DNA with the same sequence as that of the target strand is also released. At the same time, a single strand of DNA with the same sequence as the target strand (U to T) will be released, which will serve as the target to undergo the first step of the polymerase-mediated strand displacement reaction again, realizing the DDSD self-enhancement process, and the wt strand will be involved in the subsequent reaction to produce the fluorescence report.
